Up-regulated NLRP3 reduced sensitivity to gemcitabine in TNBC in vitro
TNBC cell lines, MDA-MB-231 and MDA-MB-468 were used. MDA-MB-231 and MDA-MB-468 cell lines were treated with NLRP3 agonist NSS or antagonist with the non-treated cell lines as controls. (A,C) RT-PCR assessed relative mRNA expression of NLRP3 by 2−ΔΔCq method. (B,D) After regulation of NLRP3 in both cell lines, the cells in all the three groups were exposed to 0, 1, 3, 5 nm gemcitabine for 72 consecutive hours. MTT detected the cytotoxicity in different groups after exposure to different concentrations of gemcitabine. OD value was measured at 570 nm.
