CircFMN2 up-regulates hTERT expression via sponging miR-1182
(A) HCT116 cells transfected with an miR-1182 mimic decreased the expression of the hTERT. (B) HT-29 cells transfected with an miR-1182 inhibitor increased the expression of the hTERT. (C) Bioinformatics analysis revealed the predicted binding sites between hTERT and miR-1182. (D) Luciferase reporter assay demonstrated that miR-1182 mimics significantly decreased the luciferase activity of hTERT-wt in 293T cells. (E) IHC assay indicated the expression of hTERT was significantly up-regulated in CRC tissues compared with adjacent non-tumorous tissues. (F) The qRT-PCR assay indicated the expression level of hTERT was significantly up-regulated in CRC tissues compared with adjacent non-tumorous tissues. (G) The effect of knockdown circFMN2 on protein levels of hTERT was attenuated by miR-1182 inhibitor. All tests were at least performed three times. Data were expressed as mean ± SD; **P<0.01.
